ارزیابی اثرات ضدقارچی عصاره‌های آبی، الکلی و استونی گال بلوط ایرانی (Quercus brantii Lindl. var. persica (Jaub and Spach) Zohary)

چکیده

استفاده از عصاره‌ها و اسانس‌های با منشاء گیاهی یکی از روش‌های نوین، سالم و بی‌خطر برای کنترل بیماری‌های گیاهی است. مشخص شده است ترکیبات استخراج شده از گال بلوط دارای خواص دارویی و درمانی متعدد بوده و دارای خواص ضد میکروبی بالایی نیز می‌باشند. این پژوهش با هدف ارزیابی اثرات ضد قارچی ترکیبات استخراجی از گال بلوط ایرانی (Quercus brantii var. persica) ناشی از زنبورAphelonyx persica روی تعدادی از قارچ‌های مهم بیماری‏زای گیاهی (فوزاریوم، آلترناریا، پیتیوم و فیتوفتورا) انجام شد. برای این منظور پس از تهیه عصاره آبی، استونی و متانولی از گال A. persica، طی آزمون‏های مختلف محیط‌های متناسب آگاردار و مایع دارای غلظت‌های گوناگونی از انواع عصاره‌ها تهیه و میانگین رشد و وزن پرگنه‌های قارچی و نیز اسپورزایی آنها در غلظت‌های متفاوت هر عصاره اندازه‏گیری و با شاهد مقایسه شد. نتایج نشان داد اثر ضد قارچی عصاره‌های گالA. persica وابسته به غلظت بوده و همراه با افزایش غلظت عصاره، خاصیت ضد قارچی آن‌ها افزایش می‌یابد. مقایسه میان اثر عصاره‌ها نشان داد که اگرچه عصاره‌ها در غلظت‏های مورد استفاده، خاصیت ضد قارچی معنی‏دار روی رشد و وزن پرگنه‏های قارچی نداشته‏اند، اما عصاره الکلی اسپورزایی قارچ‏ها را مهار کرد. بطوریکه عصاره الکلی در غلظت های 50 و 70 میلی‏گرم بر میلی‏لیتر و بالاتر به طور معنی‌داری موجب کاهش تولید اسپور در محیط‌های کشت قارچی شد؛ از این‌رو لازم است در مطالعات آتی در زمینه کاربرد دارویی آن تحقیقات جامع تری صورت گیرد.

One of the new and safe methods for controlling plant diseases is the use of herbal essences and extracts. It has been determined that the extracts from oak galls have multiple medicinal and therapeutic properties and also have high antimicrobial properties. The present investigation aimed to evaluate the antifungal activities of compounds extracted from oak (Quercus brantii var. persica) gall formed by Aphelonyx persica wasp against a number of important phytopathogenic fungi (Phytophthora, Pythium, Fusarium and Alternaria). After preparation of the aqueous, acetone and methanol extracts of A. persica galls, in different tests, the appropriate agar and liquid media containing various concentrations of extracts were prepared and weight average, growth and sporulation of fungal colonies were evaluated and then compared with the controls. Findings of this study demonstrated that the antifungal properties of the A. persica gall extract were concentration-dependent where its antifungal effect was increased with increasing the extract concentration. The data analysis revealed that none of the gall extracts concentrations significantly affected the growth and weight of fungal colonies, while the alcoholic extract in high doses (50 and 70 mg/ml) inhibited sporulation significantly; therefore, more comprehensive research is needed in future studies on its drug use.
